Non-Invasive Blood Pressure in Mice and Rats

Non-Invasive Blood Pressure, Rat and Mouse Measurement

The Nibp methodology consists of utilizing a tail cuff placed on the tail to occlude the blood flow. Upon deflation, one of several types of Nibp sensors, placed distal to the occlusion cuff, can be utilized to monitor the rat Bp. There are three (3) types of Nibp sensor technologies: photoplethysmography, piezoplethysmography and Volume Pressure Recording. Each formula will use an occlusion tail-cuff as part of the procedure.

1. Photoplethysmography

The first and oldest sensor type is Photoplethysmography (Ppg), a light-based technology. The purpose is to report the first appearance of the pulse while deflating the occlusion cuff or the disappearance of pulses upon inflation of the occlusion cuff. Photoplethysmography utilizes an incandescent or Led light source to report the pulse signal wave. As such, this light-based plethysmographic formula uses the light source to illuminate a small spot on the tail and attempts to report the pulse.

Photoplethysmography (Ppg) is relatively inaccurate since the readings are based solely on the amplitude of a single pulse and can only imprecisely quantum the systolic blood pressure and the heart beat. There are many limitations to a light-based technology, such as: (1) over-saturation of the Bp signal by ambient light; (2) extreme sensitivity to the rodent's movement (motion artifact); and (3) the difficulty in obtaining adequate mice blood pressure signals in dark skinned rodents (Pigmentation Differentiation). Light-based sensors also cause tail burns from close perceive and continued exposure.

Diastolic blood pressure cannot be measured by photoplethysmography since the technology records only the first appearance of the pulse. If the diastolic Bp is displayed on the photoplethysmographic instrumentation, it is only an appraisal that is calculated by a software algorithm rather than a true measurement.

The photoplethysmography formula correlates poorly with direct blood pressure measurements and is the least recommended sensor technology for Nibpe in rodents, especially mice.

2. Piezoplethysmography

The second Nibp sensor technology is piezoplethys-mography. Piezoplethysmography and photoplethysmography need the same first appearance of a pulse in the tail to report the systolic blood pressure and heart rate.

Both plethysmographic methods have similar clinical limitations. Whereas photoplethysmography uses a light source to endeavor to report the pulse signal, piezoplethysmography utilizes piezoelectric ceramic crystals to do the same. From a technical point of view, piezoplethysmography is far more sensitive than photoplethysmography since the signal from the sensor is the rate of turn of the pulse rather than just the pulse amplitude. Therefore, even very small mice with high velocity pulses will generate a adequate signal to be detected with straightforward amplifiers.

Piezoelectric sensors are more definite than light-based/Led sensors but the same plethysmographic limitations continue to yield inaccuracies in blood pressure measurements. In an independent clinical validation study conducted in 2003 at Yale University, New Haven, Connecticut, Volume Pressure Recording correlated 99 percent with direct blood pressure:

"Volume Pressure Recording is excellent. It is very definite and dependable. We performed experiments on temperature-controlled, adult rats and the non-invasive blood pressure measurements showed roughly exquisite correlation with invasive blood pressure measurements. We are very pleased with the results."

I get a lot of comments and questions regarding
nutrition. Most of them have to do with time. In
other words, population feel they are too busy to eat
5 or 6 nutritious meals a day, whether their goal
is to burn fat or build muscle.

Eat 6 Meals A Day

Meal Frequency (i.e., the amount of meals you eat each day)
could be the most prominent aspect of your nourishment program.
If you do this right you will build muscle, burn fat, and
with a proper resistance training program, be well on your
way to achieving the body of your dreams.

The thing is, most population know about it, yet few no ifs ands or buts
follow straight through with it and put it into operation because of the
reasons mentioned above.

It takes motivation, persistence, planning, and a dinky
bit of sacrifice. Sounds horrible, doesn't it? Don't worry, it's
not. But as I've stated before, nothing worth achieving comes
easy. Don't get me wrong, it is simple, it's just not easy.
But it can be pretty painless, if you have the right mind set.

Reason amount two - eating six proper, nutritious meals a day
takes work. It takes a time commitment, among other sacrifices.
And it's not just the time it takes to make the meals, or the
time to eat them. It also involves other aspects of your life.

For example, socializing, going shopping, playing sports on the
weekends, etc. If you eat six times a day, you need to eat
every 2 or 3 hours, tops. What are you going to do, sit home
all day every day so you can be near your food? Of policy not.

But you will need to plan ahead. This could mean shopping and
then cooking most of your food once a week, putting each meal
in a microwaveable container, and then carrying meals to work
with you.

You may need to invest in a high potential thermos (or two or
three) and make your beloved protein shakes to take with you
when you are going to be out and about for a while, or bringing
some protein bars with you instead.

This, more than anyplace else, is where supplementation is an
absolute life saver. I don't know how I'd get proper nourishment
day in and day out without supplements like meal exchange
powders and protein powders such as Muscle Meals and Pro-Fusion
by Muscle Link and Optimum Nutrition's Whey Protein.

I know, eating right may not be easy, but it is needful if you
truly want to perform your best body, and in the shortest amount of
time possible. If you're like most people, you already eat breakfast,
lunch and dinner. So, for starters, consider just adding two snacks
to this, preferably a protein or meal exchange shake, or bar. My
favorite bars, that recently one a contest in Men's health (for
nutrition and potential as well as taste) are Tri-O-Plex bars.

You could also consider bringing safe bet foods with you for
snacks, such as fruit, raw veggies (carrots, cauliflower, celery, broccoli
and so on). Seeds and nuts like almonds and walnuts are excellent
snacks while providing the wholesome fats we all need. Cottage cheese
and yogurt also make great snacks (preferebly the low fat variety,
so that you keep your calories in check). I commonly add a few
pineapple slices (the natural juices variety, not syrup), to my
cottage cheese.

Plan your daily (or weekly) menus. Planning is a crucial
part of success for any goal you want to achieve. Your nutrition
plan is no different.

Yes, it can be a real pain in the you know what to eat 5 or
6 times a day, but it's not impossible, and it's naturally a
part of the price you pay for a great, wholesome body. In fact,
don't even think of it as paying a price. Intead, think of how
you'l be enjoying the benefits of being wholesome and in shape.

However, with these tips I've given you, it can become easier
than you think.

If you think planning your nourishment is paying a price, wait
until you start having serious health problems from eating
improperly and being overweight - heat disease, diabetes and
the list goes on. Really, if you think it's difficult to
be wholesome and fit, try being fat and sick for a while!

Day 1 - Any fruit you would like to eat except bananas.

Day 2 - Any vegetables you want.

Day 4 - Up to six bananas, all the skim milk you want.

Day 5 - Ten to twenty ounces of beef, a can of tomatoes, 5 to 8 glasses of water.

Day 6 - All the beef and vegetables you want.

Day 7 - Consume as much unsweetened fruit juices, brown rice and fresh vegetables as you wish. Again, eat at least 1 bowl of soup during today. Don't eat any bread or drink any alcohol, or carbonated drinks.
